Венозные тромбозы этиопатогенетически связаны с наследственными и приобретенными состояниями, характеризующимися чрезмерной склонностью организма к тромбообразованию в кровеносных сосудах. Одним из типов нарушений в системе гемостаза является гестационная тромбофилия, которая может приводить к развитию венозных тромбозов во время беременности и вызывать различные осложнения в гравидарный период. Поэтому важно понимать этиопатогенез данного патологического состояния, в том числе вклад наследственных факторов. Цель. Провести анализ ассоциации 8 генетических вариантов генов системы гемостаза с отягощенным акушерско-гинекологическим анамнезом у женщин на фоне развития венозных тромбозов во время гестации. Материалы и методы. В исследование были включены 311 женщин в возрасте от 20 до 38 лет вне беременности, у которых хотя бы одна предыдущая беременность была с отягощенным акушерско-гинекологическим анамнезом, причем во время гестации были выявлены гестационные венозные тромбозы различных локализаций, и 225 женщин контрольной группы. Исследование генотипов выбранных генетических вариантов проводилось методом ПЦР в реальном времени с анализом кривых плавления. Результаты. Положительная ассоциация была выявлена для 4 генетических вариантов: 20210 G&gt;A гена F2 (генотипы GA+AA) (ОШ = 11,03 (95 % ДИ: 2,60–46,81), P &lt; 0,001); 1691G&gt;A гена F5 (генотипы GA+AA) (ОШ = 6,02 (95 % ДИ: 2,52–14,38), P &lt; 0,001); -455 G&gt;A гена FGB (генотип AA) (ОШ = 5,65 (95 % ДИ: 3,05–10,45), P &lt; 0,001) и -675 5G&gt;4G гена PAI-1 (генотип 4G/4G) (ОШ = 2,28 (95 % ДИ: 1,54–3,39), P &lt; 0,001). Заключение. Таким образом, мы установили ассоциацию 4 генетических вариантов генов плазменных факторов гемостаза с наследственной предрасположенностью к отягощенному акушерско-гинекологическому анамнезу у женщин на фоне развития гестационных венозных тромбозов.</p></abstract><trans-abstract xml:lang="en"><p>Background. One of the types of disorders in the hemostasis system is gestational thrombophilia. Therefore, it is important to understand the etiopathogenesis of this pathological condition, including the contribution of hereditary factors. Objective. To analyze the association of 8 genetic variants of the hemostasis system genes in women with gestational thrombophilia. Materials and methods. The study included 311 women aged 20 to 38 years outside pregnancy, who had at least 1 previous pregnancy with complicated obstetric and gynecological history, and gestational venous thrombosis of various localizations was detected during gestation, and 225 women in the control group. The genotypes of the selected genetic variants were studied by real-time PCR. Results. A positive association was found for 4 genetic variants: 20210 G&gt;A of the F2 gene (GA + AA) (OR = 11.03 (95 % CI: 2.60–46.81), P &lt; 0.001); 1691G&gt;A of the F5 gene (GA + AA) (OR = 6.02 (95 % CI: 2.52–14.38), P &lt; 0.001); -455 G&gt;A of the FGB gene (AA) (OR = 5.65 (95 % CI: 3.05–10.45), P &lt; 0.001) and -675 5G&gt;4G of the PAI-1 gene (4G/4G) (OR = 2.28 (95 % CI: 1.54–3.39), P &lt; 0.001). Conclusion.
