Процесс трансляции результатов отдельных доклинических исследований (ДКИ) в клиническую практику сопровождается существенным количеством неудач и сопряжен со значительными временными и финансовыми затратами. Неудовлетворительный дизайн отдельных ДКИ и отчетность, представляемая после проведения экспериментов in vivo на одной испытательной площадке, не позволяют преодолеть трансляционные барьеры и повысить воспроизводимость получаемых результатов. Актуальным становится проведение многоцентровых доклинических исследований (мДКИ) для подтверждения эффективности лекарственных средств и повышения шансов успешной трансляции доклинических результатов в клиническую практику.</p></sec><sec><title>Цель</title><p>Цель. Провести описательный анализ научной литературы, посвященной мДКИ, при изучении фармакологической активности новых лекарственных средств.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Поиск публикаций выполняли в базах данных PubMed и Google Scholar. В обзор включали публикации, доступные для поиска на 16 октября 2022 года.</p></sec><sec><title>Результаты</title><p>Результаты. Показано, что одноцентровые исследования, как правило, не позволяют точно предсказать величину эффекта, а большие размеры выборки делают оценку величины эффекта еще менее точной.</p></sec><sec><title>Выводы</title><p>Выводы. Возможно, основной целью мДКИ должна быть не количественная оценка размера эффекта на нескольких испытательных площадках, как это позиционируется в зарубежной доклинической практике, важнее оценить направленность эффекта, включая его «устойчивость», а также выявить источники гетерогенности между исследованиями, проведенными в разных центрах (лабораториях).</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Background</title><p>Background. The translating of individual preclinical studies (PCTs) results into clinical practice is accompanied by a significant number of failures and is associated with significant time and financial costs. The unsatisfactory design of individual non-clinical studies and the reports submitted after having done in vivo experiments at one laboratories do not allow overcoming translational barriers and increasing the results reproducibility. To conduct multicenter non-clinical trials is relevant to to confirm the effectiveness of medicines and increase the chances of successful translation of non-clinical results into clinical practice.</p></sec><sec><title>Objective</title><p>Objective. To analyze the scientific literature devoted to multicenter non-clinical studies in the research of the pharmacological activity of new drugs.</p></sec><sec><title>Design and methods</title><p>Design and methods. The research was done using PubMed and Google Scholar databases. The review included available publications as of October 16, 2022.</p></sec><sec><title>Results</title><p>Results. It was discovered that single-center studies did not allow predicting the magnitude of the effect accurately, and large sample sizes made the estimation of the magnitude of the effect even less precise.</p></sec><sec><title>Conclusions</title><p>Conclusions.
