Исследования в области онкологии требуют использования животных ксенографтных моделей, соответствующих принципам гуманности и рациональности. Доклинические исследования низкомолекулярных ингибиторов, противоопухолевых моноклональных антител, вакцин, клеточных препаратов, в том числе CAR-T клеток, нуждаются в прижизненном контроле динамики роста опухоли. Широко применяемая люциферазная детекция биолюминесценции ксенографтов требует введения субстрата, однако инъекции менее гуманны, более трудоемки, дорогостоящи, стрессируют животное. Альтернативой люциферазной биолюминесценции могут быть флуоресцентные белки дальне-красного спектра, визуализация эмиссии которых в экспериментальных моделях in vivo упростит манипуляции с животными и увеличит доступность детекции.</p></sec><sec><title>Цель</title><p>Цель. Сравнить эффективность детекции динамики роста опухолевых ксенографтов in vivo в мышиных моделях с инъекцией опухоли шейки матки, применяя доступную линейку флуоресцентных дериватов красного белка одной клады Katushka с распространенной в in vivo исследованиях люциферазой Renilla.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Ксенографтные модели NSG-SGM3 мышей были получены подкожным введением модифицированных клеток линии HeLa, экспрессирующих флуоресцентные белки-репортеры Katushka, Katushka2S, TurboRFP, TurboFP650 и люциферазу Renilla. Исследование и оценку флуоресцентного и биолюминесцентного сигнала опухоли проводили на 7, 14, 21 сутки после введения ксенографта.</p></sec><sec><title>Результаты</title><p>Результаты. Спектральные характеристики и детекция эмиссии белков-репортеров Katushka и Katushka2S выделяют данные маркеры среди всей линейки исследуемых белков дальне-красного спектра.</p></sec><sec><title>Заключение</title><p>Заключение. Детекция флуоресцентного сигнала дальне-красных белков-репортеров Katushka и Katushka2S может служить полноценной альтернативой люциферазной биолюминесценции фермента Renilla в экспериментальных моделях in vivo в рамках иммунотерапевтических исследований.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Background</title><p>Background. Research in oncology assumes establishment and usage of xenograft animal models, meeting the requirements of humanity and rationality. Although praised as promising field of research, preclinical studies of low-molecular-weight inhibitors, antitumor monoclonal antibodies, vaccines, cellular therapy products, CAR-T cells entail continuous control of tumor growth dynamics. Luciferase detection of bioluminescence requires injection of a reporter substrate. However, injections not only are laborious, time-consuming and expensive, but are also stressful for animals. Thus, a vast variety of new visualization methods is employed, including proteins of the far-red spectrum.</p></sec><sec><title>Objective</title><p>Objective. The study aimed to compare detection efficiency of tumor growth dynamics in mice models of cervical cancer, applying a commercially available line of fluorescent red protein derivatives of Katushka clade and Renilla luciferase, that is commonly used in in vivo studies. </p></sec><sec><title>Design and methods</title><p>Design and methods. Xenograft mice models were derived injecting modified HeLa cell line, that expresses fluorescent reporter proteins: Katushka, Katushka2S, TurboRFP, TurboFP650 and Renilla enzyme. </p></sec><sec><title>Results</title><p>Results. Spectral properties and emission wavelength of far-red fluorescent protein Katushka and Katushka2S outlines these markers within RFP derivatives lineage as outstanding instrument for in vivo tumor visualization.</p></sec><sec><title>Conclusion</title><p>Conclusion.
