Несмотря на функциональную множественность и сложность действий TNF- α, его роль в центральной нервной системе при развитии различных патологических состояний в организме до сих пор не понята.</p></sec><sec><title>Цель</title><p>Цель. Изучить динамику TNF-a и его рецепторов в головном мозге мышей с нокаутом по урокиназе (uPA-) при злокачественном росте на фоне хронической нейрогенной боли (ХНБ).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Работа выполнена на мышах обоего пола двух линий: C57BL/6 (n = 80) и C57BL/6-PlautmI.IBug-ThisPlau6FDhu/GFDhu (n = 56). В основных группах животных ХНБ моделировали путем двусторонней перевязки седалищных нервов, через 2 недели под кожу спины перевивали меланому В16/ F10. Группа сравнения — ложно оперированные животные с перевивной меланомой. Контрольные группы — ложно оперированные животные и животные с ХНБ. На 21 сутки роста меланомы мышей декапи-тировали, в церебральной ткани методом иммуноферментного анализа определяли содержание TNF- α, sTNF- α R1 и sTNF- α R2.</p></sec><sec><title>Результаты</title><p>Результаты. В мозге самцов uPA- регистрировался высокий уровень TNF- α (в 5,6 раза) и низкий — TNF- α R1 (в 2,1 раза), у самок — изменений не было. Под влиянием ХНБ содержание TNF- α в мозге становилось одинаковым у самцов и самок мышей, при этом у мышей uPA- уровень TNF- α был более чем в 2,0 раза больше, чем у мышей с нормальным геномом. Основу стимулирующего действия ХНБ на злокачественный процесс у мышей uPA- составили: у самок — увеличение церебральной концентрации TNF- α R2 (в 1,7 раза; р &lt; 0,05), у самцов — редукция TNF- α (в 2,2 раза).</p></sec><sec><title>Заключение</title><p>Заключение. Специфика фибринолитического статуса мышей определяет течение злокачественного процесса подкожно перевитой меланомы на фоне боли, один из механизмов этого явления — изменение функционирования системы TNF- α головного мозга, зависящее от пола животного.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Background</title><p>Background. Despite the functional multiplicity and complexity of TNF- α effects, its role in the central nervous system in the development of various pathological conditions of the body is still poorly understood.</p></sec><sec><title>Objective</title><p>Objective. Analyzing the dynamics of TNF- α and its receptors in the brain of urokinase (uPA-) gene-knockout mice with malignant growth in presence of chronic neurogenic pain (CNP).</p></sec><sec><title>Design and methods</title><p>Design and methods. The study included male and female mice of two strains: C57BL/6 (n = 80) and C57BL/6-PlautmI.IBug-ThisPlau6FDhu/ GFDhu (n = 56). A CNP model was created in animals of the main groups by the bilateral sciatic nerve ligation; 2 weeks after it, B16/F10 melanoma was transplanted under the skin of the back. The comparison group — sham operated animals with transplanted melanoma. Control groups — sham operated animals and animals with CNP. On day 21 of the melanoma growth, the mice were decapitated, and levels of TNF- α, sTNF- α R1 and sTNF- α R2 were determined in the cerebral tissues by ELISA.</p></sec><sec><title>Results</title><p>Results. High levels of TNF- α (by 5.6 times) and low TNF- α R1 (by 2.1 times) were registered in the brain of uPA- males, while no changes were registered in females. Under the influence of CNP, brain levels of TNF- α became similar in male and female mice; TNF- α levels in uPA- mice were twice higher than in animals with the normal genome. The CNP stimulating effect on the malignant process in uPA- mice was based: in females — on an increase in the cerebral concentration of TNF- α R2 (by 1.7 times, p&lt;0.05), in males — on TNF- α reduction (by 2.2 times).</p></sec><sec><title>Conclusion</title><p>Conclusion.
