Нами ранее показано, что пальмитолилированный пептид 612-627 (Pal), структурно соответствующий третьей внутриклеточной петле рецептора тиреотропного гормона (ТТГ), активирует аденилатциклазную сигнальную систему в тироидальных мембранах in vitro и стимулирует продукцию тироксина щитовидной железой in vivo. Цель настоящей работы состояла в исследовании дозозависимости стимулирующего влияния пептида 612-627 (Pal) на уровень тиреоидных гормонов при его интраназальном и внутримышечном введении крысам. Пептид вводили однократно самцам крыс Wistar в дозах от 45 до 900 мкг/кг при интраназальном введении и в дозах от 150 до 1350 мкг/кг при внутримышечном введении. До введения и в течение 6 ч после него оценивали динамику изменений уровня свободного (fT4) и общего тироксина (tT4) и общего трийодтиронина (tT3). Показано, что при обоих путях введения пептид 612-627 (Pal) дозозависимо повышал уровень fT4 и tT3 В то же время интраназальный путь был более эффективным, поскольку значение ED50 и максимальный стимулирующий эффект на продукцию fT4 при интраназальном введении пептида составили 87 мкг/кг и 38 %, в то время как при внутримышечном - 275 мкг/кг и 25 %, соответственно. Полученные данные указывают на способность пептида 612-627 (Pal) эффективно стимулировать гормонопродуцирующую функцию щитовидной железы при его интраназальном введении крысам в диапазоне доз 225-450 мкг/кг.</p></abstract><trans-abstract xml:lang="en"><p>Background. Search and development of new regulators of functional activity of the hypothalamic- pituitary-thyroid axis is one of actual problems in endocrinology and pharmacology. We have previously shown that palmitoylated peptide 612-627 (Pal) structurally corresponding to the third intracellular loop of receptor of thyroid-stimulating hormone (TSH) activates the adenylyl cyclase signaling system in thyroidal membranes in vitro and stimulates the production of thyroxine by the thyroid gland in vivo. Objective. The aim of this work was to study a dose-dependency of the stimulating effect of peptide 612-627 (Pal) on the level of thyroid hormones in its intranasal and intramuscular administration into rats. Design and methods. The peptide was administered once to Wistar male rats at the doses from 45 to 900 pg/kg when administered intranasally and at the doses from 150 to 1350 pg/kg when administered intramuscularly. Prior to administration and for 6 h after administration the changes of the levels of free (fT4) and total thyroxine (tT4) and total triiodothyronine (tT3) were evaluated. Results. It is shown that peptide 612-627 (Pal) in both routes of administration in a dose- dependent manner increased the levels of fT4 and tT3 At the same time, the intranasal route was more effective as ED50 value and the maximal stimulating effect on fT4 production when peptide administered intranasally was 87 pg/kg and 38 %, while in the case of intramuscular route - 275 pg/kg and 25 %, respectively. Conclusion.
