Однако, в большинстве известных стрептозотоциновых моделей развивается диабет с выраженной инсулинопенией без ожирения, дислипидемии и инсулинорезистентности, что, как правило, не характерно для сахарного диабета 2-го типа. Нами предложена модель диабетической нефропатии при сахарном диабете 2-го типа, индуцированном внутрибрюшинным введением половозрелым самцам крыс стока Wistar последовательно с интервалом в 15 минут растворов никотинамида (230 мг/кг) и стрептозотоцина (65 мг/кг). Предварительно крысам выполнялась правосторонняя нефрэктомия с назначением в дальнейшем на 5 недель высокожирового питания, содержащего говяжье сало. Согласно полученным данным, у крыс с диабетом в описываемой модели по сравнению с контрольной группой, также подвергшейся геминефрэктомии, но находившейся на стандартном лабораторном рационе без введения стрептозотоцина и никотинамида, развиваются избыточной вес, умеренные гипергликемия, инсулинорезистентность и дислипидемия. На 30-й неделе эксперимента развитие диабетической нефропатии подтверждено значимым увеличением экскреции альбумина с мочой, снижением клиренса креатинина, а также наличием характерных морфологических почечных признаков по результатам световой и электронной микроскопии, что позволяет рассматривать предложенную модель у крыс в качестве валидного инструмента для моделирования относительно ранних стадий диабетической нефропатии при диабете 2-го типа, наблюдаемых у пациентов.</p></abstract><trans-abstract xml:lang="en"><p>Non-genetic rat models of diabetic nephropathy are most commonly reproduced by using streptozotocin. However, administration of high doses of streptozotocin might have significant disadvantages such as severe hyperglycemia due to insulinopenic condition. Nevertheless, certain significant metabolic factors besides hyperglycemia, such as insulin resistance, obesity, dyslipidemia, and hypertension plays a crucial role in the onset and progression of diabetic nephropathy in type 2 diabetes mellitus. Hereby, we have described a novel model of type 2 diabetes and nephropathy in rats that is characterized by morphofunctional changes similar to relatively early stages of human diabetic nephropathy. Starting at 3 weeks after right-side nephrectomy, adult male Wistar rats were fed for 5 weeks the high-fat diet containing beef tallow and then successively received nicotinamide (230 mg/kg) and streptozotocin (65 mg/kg) intraperitoneally in 15-min interval. Control nondiabetic uninephrectomized rats received vehicle and were fed normal chow. With described model we observed the development of mild hyperglycemia, overweight, slight significant increase in insulin resistance, and dyslipidemia which resulted in a gradual decrease in renal function up to significant reduced creatinine clearance and elevated albuminuria, and confirmed diabetic nephropathy by validated electron and light microscopic morphological criteria at week 30 of the study.</p></trans-abstract><kwd-group xml:lang="ru"><kwd>крысы</kwd><kwd>негенетическая модель</kwd><kwd>сахарный диабет 2-го типа</kwd><kwd>диабетическая нефропатия</kwd><kwd>rats</kwd><kwd>non-genetic model</kwd><kwd>type 2 diabetes mellitus</kwd><kwd>diabetic nephropathy</kwd></kwd-group></article-meta></front><back><ref-list><title>References</title><ref id="cit1"><label>1</label><citation-alternatives><mixed-citation xml:lang="ru">Шестакова М.В, Дедов И.И.
