Внеклеточный матрикс играет ключевую роль в поддержании гомеостаза и регенерации костной ткани, определяя условия дифференцировки и межклеточных взаимодействий. Нативный внеклеточный матрикс представляет собой сложную трехмерную сеть структурных белков, гликопротеинов и протеогликанов, обеспечивающих механическую поддержку клеток и передачу регуляторных сигналов. Традиционные 2D-системы культивирования не позволяют в полной мере воспроизводить сложную организацию клеточного микроокружения, поэтому актуальной задачей является разработка трехмерных систем для изучения процессов дифференцировки и ремоделирования костной ткани in vitro.Цель. Оценка биосовместимости гидрогеля на основе экстракта миомы матки и его влияния на остеогенную дифференцировку остеобластов в моно- и сокультуре с эндотелиальными клетками.Материалы и методы. Экстракт миомы матки выделяли по модифицированной методике, разработанной для получения Matrigel® из саркомы Энгельбрета-Хольма-Сварма мышей. Клеточную суспензию остеобластов смешивали с экстрактом миомы матки на холоде, затем смесь инкубировали при 37 °C для образования гидрогеля. Остеобласты культивировали в гидрогеле в виде моно- и сокультуры с эндотелиальными клетками в остеогенной среде. Жизнеспособность и морфологию клеток оценивали методом прижизненной микроскопии. Минерализацию внеклеточного матрикса определяли методом окраски ализариновым красным.Результаты. Установлено, что гидрогель на основе экстракта миомы матки обладает высокой биосовместимостью в отношении остеобластов – поддерживает их жизнеспособность, способствует дифференцировке в остеоцитоподобные клетки и усиливает минерализацию внеклеточного матрикса. В сокультуре остеобластов с эндотелиальными клетками эффект усиливался, что указывает на важную роль межклеточных взаимодействий.Заключение. Гидрогель на основе экстракта миомы матки представляет собой перспективную 3D-модель культивирования для изучения процессов остеогенной дифференцировки остеобластов.</p></abstract><trans-abstract xml:lang="en"><p>Background. The extracellular matrix plays a key role in maintaining homeostasis and bone tissue regeneration by defining the conditions for cell differentiation and intercellular interactions. The native extracellular matrix is a complex three-dimensional network of structural proteins, glycoproteins, and proteoglycans that provide mechanical support to cells and transmit regulatory signals. 2D-culture systems do not reproduce the complex organization of the cellular microenvironment; therefore, the development of 3D-systems for bone tissue differentiation and remodeling in vitro remains an important challenge. Objective. To evaluate the biocompatibility of the hydrogel based on a tissue extract of uterine leiomyoma and its potential to regulate osteogenic differentiation and mineralization of osteoblasts in mono- and co-culture with endothelial cells. Materials and Methods. The tissue extract of uterine leiomyoma was obtained using a modified protocol originally developed for producing Matrigel®. Osteoblasts were cultured within the hydrogel in mono- and co-culture with endothelial cells under osteogenic conditions. Cell viability and morphology were assessed by live-cell microscopy. Mineralization of the extracellular matrix was evaluated using Alizarin Red S staining. Results. The hydrogel based on tissue extract of uterine leiomyoma demonstrated high biocompatibility toward osteoblasts, supporting their viability, promoting differentiation into osteocyte-like cells, and enhancing extracellular matrix mineralization. In co-culture with endothelial cells, these effects were further enhanced, indicating the important role of intercellular interactions. Conclusion.
