Руксолитиниб является селективным ингибитором Янус-киназ 1 и 2, подавляющим JAK/ STAT-зависимую сигнализацию, вовлеченную в воспаление, иммунный ответ и клеточную пролиферацию. Помимо системного применения, интерес представляет ингаляционный путь введения препарата, который потенциально позволяет повысить его локальную экспозицию в легких и снизить системное воздействие. Целью исследования стала разработка технологии радиоизотопного мечения руксолитиниба углеродом-11 для последующего изучения его биораспределения при различных путях введения.Материалы и методы. N-[11C]-метилруксолитиниб получали одностадийной реакцией метилирования руксолитиниба на полностью автоматизированном самодельном модуле. Очистку продукта осуществляли методом твердофазной экстракции, радиохимическую чистоту и подлинность подтверждали методом радио-ВЭЖХ. Биораспределение изучали ex vivo на здоровых крысах-самцах Wistar при внутривенном и ингаляционном введении радиолиганда. Накопление радиоактивности в органах оценивали через 5, 10, 40, 60 и 80 мин. после введения.Результаты. N-[11C]-метилруксолитиниб получен с радиохимическим выходом 40 % (при пересчете на [11C]CH3I), радиохимической чистотой ≥97 % и мольной активностью 6‒10 ГБк/мкмоль. В пилотном исследовании установлено, что при ингаляционном введении через 5 мин. после введения наблюдалась тенденция к более высокому накоплению радиоактивного лиганда в легких по сравнению с внутривенным введением. Одновременно при внутривенном пути введения в этой же точке отмечалась тенденция к более высокому накоплению в печени. Однако уже к 10-й минуте существенные различия в биораспределении между двумя путями введения нивелировались.Заключение. В настоящей работе впервые описан радиосинтез N-[11C]-метилированного аналога руксолитиниба, а также проведена сравнительная оценка его биораспределения при системном и ингаляционном путях введения.</p></abstract><trans-abstract xml:lang="en"><p>Introduction. Ruxolitinib is a selective inhibitor of Janus kinases 1 and 2, suppressing JAK/STAT-dependent signaling involved in inflammation, immune response, and cell proliferation. In addition to systemic use, inhalation administration is of interest, potentially increasing local lung exposure and reducing systemic exposure. This study aimed to develop a method for radioisotope labeling of ruxolitinib with carbon-11 to investigate its biodistribution via different administration routes.Materials and Methods. N-[11C]-methylruxolitinib was obtained by a one-step methylation reaction of ruxolitinib using a fully automated, home-made module. The product was purified by solid-phase extraction, and radiochemical purity and identity were confirmed by radio- HPLC. Biodistribution was studied ex vivo in healthy male Wistar rats after intravenous and inhalation administration of the radioligand. Radioactivity accumulation in organs was assessed at 5, 10, 40, 60, and 80 minutes after administration.Results. N-[11C]-methylruxolitinib was obtained with a radiochemical yield of 40 % (when calculated as [11C]CH3I), a radiochemical purity of ≥97 %, and a molar activity of 6‒10 GBq/μmol. A pilot study found that with inhalation administration, a tendency toward higher radioligand accumulation in the lungs was observed 5 minutes after administration compared to intravenous administration. In contrast, with intravenous administration, a tendency toward higher accumulation in the liver was observed at the same time point. However, by the 10th minute, significant differences in biodistribution between the two routes of administration had disappeared.Conclusions.
