Мембранно-ассоциированный белок теплового шока Hsp70 (mHsp70) избирательно экспрессируется в опухолевых, но не в нормальных клетках. Спектр функций mHsp70 до сих пор не до конца изучен. Цель — исследование роли mHsp70 в миграции клеток глиобластомы. Материалы и методы. В работе использовали клеточные линии С6 глиомы крысы, U251 и T98G глиобластомы человека, положительные по mHsp70. Каждую клеточную линию сортировали на две субпопуляции — с высокой (mHsp70+) и низкой (mHsp70-) экспрессией белка. Вклад mHsp70 в миграцию оценивали с помощью теста на зарастание раны, ручного трекинга единичных клеток и метода Transwell. В работе также иссле довали влияние ингибиторов белка PES и JG-98 на клеточную подвижность. Для выявления возможных белковых партнеров mHsp70, регулирующих клеточную подвижность, проводили протеомный анализ липидных рафтов клеток T98G. Результаты. Субпопуляции mHsp70+ обладают более высокой скоростью движения (по данным ручного трекинга) и приводят к полному зарастанию раны за более короткий промежуток времени по сравнению с mHsp70-. Применение ингибиторов PES и JG-98 способствует снижению скорости движения, а также уменьшению количества инвазирующих клеток, при этом наи больший эффект наблюдается для субпопуляций mHsp70+. Протеомный анализ липидных рафтов клеток T98G выявил связь между mHsp70 и белками, участвующими в ремоделировании цитоскелета, внеклеточного матрикса, адгезии и миграции. Заключение. mHsp70 участвует в миграции клеток глиобластомы и может быть использован в качестве мишени для тераностики злокачественных новообразований.</p></abstract><trans-abstract xml:lang="en"><p>Background. Membrane-associated heat shock protein Hsp70 (mHsp70) is selectively expressed in tu mor, but not in normal cells. The mHsp70 functions are not fully understood. Objective: to study the role of mHsp70 in glioblastoma cell migration. Design and methods. mHsp70-positive rat glioma C6 and human glioblastoma U251 and T98G cell lines were used. Each cell line was sorted into two subpopulations: with high (mHsp70+) and low (mHsp70-) protein expression. The contribution of mHsp70 to migration was as sessed using a wound-healing assay, manual single-cell tracking, and the Transwell analysis. We also exam ined the effect of Hsp70 inhibitors PES and JG-98 on cell motility. To identify potential protein partners of mHsp70 that regulate cell motility, proteomic analysis of lipid rafts of T98G cells was performed. Results. mHsp70+ subpopulations have a higher mean speed (according to manual tracking) and lead to complete wound healing in a shorter period of time compared to mHsp70-. The use of PES and JG-98 inhibitors helps to reduce the speed of movement, as well as the number of invasive cells, with the greatest effect observed for mHsp70+ subpopulations. Proteomic analysis of T98G cells lipid rafts revealed a relationship between mHsp70 and proteins involved in cytoskeleton and extracellular matrix remodeling, adhesion and migration.
